Abstract: Astatic soda ponds of the Carpathian Basin are light-limited due to their high suspended matter content and/or chromophoric dissolved organic matter content. This and their special chemical composition, furthermore their high nutrient content, make these habitats unique worldwide. Due to their characteristic physico-chemical properties, microbial communities living in these ponds are also unique. This study aimed to obtain a more detailed view about the structure and function of microorganisms with high-resolution genomic tools in the case of a soda pond prior desiccation, Büdös-szék pond near Szabadszállás in November 2012. For taxon identification, the V3-V4 region of the 16S rRNA gene was analyzed on a Roche GS Junior DNA sequencer. For the functional analysis, a shotgun metagenomic approach was applied on an Ion Torrent PGM genetic analyzer. Results showed a taxonomically and functionally complex microbial community structure dominated by members of the phyla Proteobacteria, Actinobacteria, and Bacteroidetes with the remarkable contribution of two uncultured Actinomicrobiaceae genera and the genera Gracilimonas, Belliella, Hydrogenophaga, Roseovarius and their relatives. Functional genomic analysis revealed various modes of adaptation to the alkaline environment and to the desiccating character of the studied habitat, which were present in the community gene pool; furthermore, the importance of the serine-pathway in the carbon cycle of this environment.
