Abstract: In the second chapter of the review the methods of detecting the pathogenic unicellular Giardia lamblia in water are described. Since the concentration of the Giardia cysts in the contaminated water may be very low, large quantities (min. 100 gals =380 litres) must be filtered for successful detection. For this purpose membrane filter (Fig. 1) and orlon filter (Fig. 2) techniques have found wider application. The membrane filter tested by the author is recommended for use in Hungary. Enrichment will improve the efficiency of the filter techniques. Zinc-sulphate flotation known from parasitological practice is simple in comparison with saccharose enrichment and is suggested for routine water analyses. The immunofluorescent technique of Giardia detection has only a future potential as yet. The Giardia cyst capable of decysting are noted to present a contamination hazard, so that the determination of the viability of the cysts detected in water constitutes and important part of the Giardia tests. The in-vitro viability tests are too laborious for routine work. Familiarity with the Giardia lamblia testing techniques, the successful isolation of the cysts from water aro prerequisites to preventing contamination by this pathogen. The methods of control will be described in a subsequent chapter.
