The integrity of the epithelial cell layers is maintained by intercellular junctional complexes. One of these complexes is the tight junction (TJ) having two functions: by acting as a barrier, the TJ regulates the passage of ions, water and macromolecules through paracellular spaces, while its fence function maintains the cell polarity. The integral membrane proteins constituting the TJ strands provide targets and/or receptors for factors expressed and/or released by viruses, bacteria, or parasites. The role of integral membrane proteins in etiology of periodontitis is not yet known. The purpose of the study was to develop a method suitable for quantification of occludin and claudin (CLDN) 1, 4, 7, tight-junction-associated proteins in healthy human gingival epithelium. Subjects and methods: The expression of integral membrane proteins of TJ was analyzed in histological- j ly evaluated, healthy gingival samples obtained from five young patients prior to surgical removal of impacted mandibular third molar. The amount of Occludin and Claudins tight junction protein was quantified in different : epithelial cell layers. The digital images captured from the stained samples were optimized and converted by Adobe Photoshop software to grayscale TIFF files. The semiquantitative analysis of Occludin expression was performed by using ImageQuant software (Molecular Dynamics). The background was eliminated by a thres hold setup. On the images in each cell layers we set up 10 randomly selected polygonal areas with equal | size, and average pixel intensities of each polygons j were determined. The mean and standard deviation of grayscale intensities were calculated and plotted on graph. The Statistica 6.1 software (StatSoft Inc., Tulsa, USA) was used for statistical analysis. The statistical significance was determined by ANOVA probe and Tukeys post-hoc test. Results: The expression of Occludin increased significantly in the basal cell layer of gingival epithelium, and the expression of CLDN1, CLDN4 and CLDN7 were elevated significantly in spinous cell layer of gingival epithelium. This semiquantitative analysis confirmed the results recorded by two independent observers. 99. évf. 6. sz. 2006. Conclusion: This method provides a tool for the semiquantitative evalution of the integral membrane proteins of TJ . Department of Dentistry and Oral Surgery, University of Szeged, Hungary One risk factor of destructive periodontitis is the increasing mobility, because that may contribute to the progression of inflammation, and change in the occlusion - which can be the result of the tooth mobility - may negatively influence the success of periodontal treatment. If periodontal support of teeth cannot compensate the primer or secondary traumatic occlusion, the treatment has a bad prognosis. Periodontitis and its consequences (e.g. tooth loss, migration, drifting) change the occlusion, and the possible correction are: adjustment of occlusion, splinting, making fixed restoration. Complex periodontal rehabilitation - including orthodontic treatment - have increasing importance today. The orthodontic treatment is suitable for replacing certain surgical interventions as lengthening crown or augmentation of jaws. Critical point is how to support the result of orthodontic treatment, practically the retention phase of teeth. In case of teeth with chronic periodontal inflammation it is not only important to prevent the movement, but with splinting of teeth we should build up a stabile occlusion. Dentists have to keep in mind that the retainer cannot impede the home care and professional oral hygiene. In this lecture authors present some types of orthodontic treatment in periodontitis patients and show how to keep “hopeless” teeth. They also demonstrate special aspects of retention treatment phase.
